Veterinary Diagnostic Laboratory


The Veterinary Diagnostic Laboratory (VDL) of the University of Illinois, College of Veterinary Medicine is pleased to announce the introduction of molecular diagnostic testing based on the highly sensitive real-time reverse transcription polymerase chain reaction (qRT-PCR) methodology for the detection of bovine viral diarrhea virus (BVDV) and porcine respiratory and reproductive syndrome virus (PRRSV).

The BVDV qRT-PCR detects and differentiates between the conserved 5' untranslated region (5' UTR) of BVDV types I and II genomes. It can detect both acutely infected and persistently infected (PI) animals. For diagnosis of acute infections, submit tissues (mesenteric lymph node, thymus, intestinal mucosa). For best results with detection of BVDV PI animals, submit fresh serum (1 ml) or an ear punch from the base of the pinna. All specimens should be kept cold during shipment. Samples must be received by 9:00 a.m. Wednesday for results Thursday afternoon. Both BVDV types I and II qRT-PCR will be run on each sample. The cost is $33.00 per serum sample and $35.00 per tissue sample.

The PRRSV qRT-PCR multiplex assay detects the conserved 3' untranslated region of the North American or European strains of the viral genome. qRT-PCR strain differentiation of multiplex- positive samples can be performed for both North American (type 2) and European (type 1) strains of PRRSV as requested. For best results submit fresh semen (5 ml raw or 50 ml extended), serum (1 ml) or tissue (lung, bronchial lymph nodes, tonsils). All specimens should be kept cold during shipment. Testing dates for serum/ear blood samples will be every day but Wednesdays with same-day results available by evening if samples are submitted by noon. Testing dates for semen and tissues will be Tuesday (samples submitted by Monday 4:30 PM) and Friday (samples submitted by Thursday 4:30 PM) each week with a 24-hour turn around time. The cost is $36.00 per serum, blood or plasma sample, $37.00 per pooled sera, blood or plasma samples, $38 per tissue, semen, or extended semen samples, and $39.00 per pooled tissue, pooled semen samples, or pooled extended seman samples.

PRRSV sequencing of the ORF5 region is available for samples that are found to be positive by qRT-PCR. Alignment to reference strains is included. Cost is $122.00 with a 10-day turn around time.

PRRS ORF5 Sequence Alignment of current to previous cases reference strains is available with a 10-day turn around time. Cost is $60.

Porcine circovirus type 2 PCR is now offered through the UI VDL Molecular Laboratory. The polymerase chain reaction assay (PCV-2 PCR) detects only the type 2 and not the type 1 strain. It is a highly sensitive and specific assay for the viral genome. Appropriate samples include serum (2 ml) or tissue (lung, tracheal-bronchial lymph nodes, small intestines). All specimens should be kept cold during shipment; tissues may be frozen. The assay will be run on Wednesdays (samples must be received by Tuesday 3:00 PM) with results reported by the following Thursday afternoon. The cost is $30.00 per serum sample, $32.00 per pooled sera sample, and $35.00 per tissue or semen sample.
